Yes, there was a multiplayer mode in the game originally, and we know that from multiple interviews. They tried to have Mario and Luigi running around together on some test maps. They tried multiple camera angles, but it just didn't work. So, he was scrapped hard. Luigi wasn't in the retail release of the game's files, he was found in the source code. But there was no multiplayer functionality found in the source code.

And another reason was because the N64 didn't have the power to have two controllable characters at once.
