I'm choosing GCN Mushroom Bridge. This might remind you of some BCP versions of retro courses...

Lap 1: It's the same as the GCN version, except both the Warp Pipe near the starting line and the sand shortcut are blocked off.
Lap 2: Once you get about a quarter of the way through the next lap, the bridge starts collapsing. You now are forced to take the sand shortcut (it's not offroad, luckily) and instead of it leading back on to the main path, you go into the village and eventually end up coming out of the Warp Pipe near the starting line.
Lap 3: The same as Lap 1, except backwards. You can glide across the broken bridge or trick off the sinking vehicles.

I've done a lot of courses in the BCP style: where at Lap 2 or 3 there's an alternate route that makes you go backward for the rest of the race

I have another one: N64 Bowser Castle.
At Lap 2, Marty's Cage opens up and you have to go through it. It turns out to lead to a factory set up on the castle turrets where Bowser produces Bob-ombs, Mechakoopas and the like. You exit out a tower-like structure. At Lap 3, the whole castle takes flight like in Paper Mario 64, and you do the lap backwards.

i've got another one
GBA Riverside Park
Lap 1: Normal except the first jump leads in to an open river which spans until the first Ptooie section
Lap 2: the first turn is blocked off and a shortcut opens up. after the shortcut, it's the same as Lap 1
Lap 3: the cave is flooded and there's Cheep Cheeps in it. aside from that, same as Lap 1

SNES Vanilla Lake 1 but you can enter in the water, the ice cubes are now freezes and in the right side of the Track there's a cave...

Another one:
GCN Daisy Cruiser but at the final lap the cruiser will crash and sink into the ocean and it will rotate 90 degrees and will be a underwater and anti-gravity section.

This topic is dead now, so thats why i have another one: 3DS Music Park (Melody Motorway) but at the final lap Hammer Bros will throw hammers at the track on the beat. (Gotta love the Mario Party references.)

Twist: after the first set of items, it's anti-gravity
then things start to get complicated like that big cheep cheep might hit a racer and VERY VERY Sharp turns.
